Description
Traditional methods for determining simple sugars and larger saccharides are typically slow with only modest resolution. However, recent developments in HPLC column chemistry have produced high-resolution options for polar compounds like sugars and saccharides. This presentation will extend earlier work on simple sugars to the larger oligo- and poly-saccharides, using a similar approach to characterize beer during and after fermentation. The combination of a fructan-like stationary phase with an evaporative light scattering detector (ELSD) allows the profiling of sugars up to about DP20. Operating conditions can be adjusted either to enhance resolution or reduce analysis time. Sample preparation also plays a key role as the profile is affected by the solubility of the larger polysaccharides. We will discuss the optimization of these factors and show applications at various stages in the production process, from wort to fermented beer.
