A Closer Look at the Design Requirements and Challenges of Point-of-care Tests for Disease Detection

Description

The field of sensor platforms, both laboratory-based and portable, is rapidly growing, and more innovative approaches are constantly being developed. This innovation is, in part, fueled by devastating diseases and pandemics throughout history. Approaches for designing, fabricating, and validating point-of-care (POC) diagnostic tests will be discussed. Furthermore, common challenges that result from miniaturization of sensor platforms and important lessons learned from sample pretreatment will be presented. This work will draw upon two examples (COVID-19 and tuberculosis) from our current research: (1) The current global COVID-19 outbreak has exposed significant shortcomings in our ability to accurately test for the presence of SARS-CoV-2 and deliver diagnostic tests on a scale to sufficiently understand the true impact on the population. Today’s COVID-19 laboratory-based tests are simply too slow and plagued with false negative test results delaying life-saving treatment and/or preventing further spread of the virus. Therefore, an urgent need for a highly sensitive and specific POC diagnostic test exists, which provides clinicians with test results in minutes. Equipped with such a test, healthcare providers could immediately begin optimal treatment plans for those infected with the SARS-CoV-2 and minimize the spread of the virus by initiating self-quarantine measures. (2) According to the World Health Organization, tuberculosis remains one of the world’s deadliest diseases with over 1.5 million death in 2018. The disease is most prevalent in regions of the world plagued by poor clinical infrastructure. However, if detected early Tuberculosis can be cured. Therefore, a low-cost POC test capable of detecting disease markers at early stages of infection is urgently needed to initiate treatment.
