High-throughput Sampling and Mass Spectral Analysis with Nanopipette-based Probes

Description

Nanopipettes are robust probes extensively used for microscopy techniques. They have been employed in our lab previously for selective analyte extraction and to study chemical reactions in local environments through mass spectrometry. The study presented here describes development of a nanopipette-based probe for simultaneous sampling and chemical analysis by nanoelectrospray ionization mass spectrometry (nanoESI-MS). A bench-top robotic arm is utilized for automated sampling through confined areas. Nanometer range tip diameters of nanopipettes for sampling can allow local probing of analytes and chemical differences in aqueous solution. In addition, small size of the nanopipette provides advantageous characteristics such as ability to aspirate small volumes of analyte solutions and higher sensitivity; in addition to advantages provided by nanoESI. Furthermore, automation provides a high-throughput method that enables efficient sampling and analysis of multiple samples in a go. Implementation of nanopipette-based probe can serve as a tool to study chemical heterogeneity in environmentally and biologically important samples, with high spatial resolution and sensitivity.
