Modern Raman Imaging: From Single Cell to Whole Animal

The last 25 years has seen Raman spectroscopy (RS) advance from a relatively slow imaging technique, that took a day or more to image a whole pharmaceutical tablet, to an accepted rapid and powerful technique capable of imaging large areas in seconds. The inherently low scattering cross section of RS, as well as its diffraction-limited lateral resolution, has been overcome by new Raman microscopy techniques. In some cases, video rate Raman imaging is now becoming a reality. Three dimensional confocal Raman imaging is now common. At the same time, utilizing the combined technique of Raman and AFM through the application of Tip Enhanced RS (TERS), beautiful Raman images on a nanometric scale are now possible. These capabilities have become possible not only through advances in Raman components and spectrometer design, but also smart selective sampling and multivariate analysis software. Recently the use of novel Raman reporter molecules with SERS or isotopic tags has allowed both increases in specificity and sensitivity. This talk will review the current tools employed in Raman imaging, recent advances, and examples of applications that these new methods have enabled. Applications illustrated will include large area mapping of pharma samples, invivo imaging of cancer through the intact skull of small animals, DNA imaging and sequencing, and nanometric imaging for the characterization of 2D materials. The enabling Raman technologies reviewed will include Raman microscopy, Spatially Offset RS (SORS), Stimulated RS (SRS), particle correlated RS (PCRS) and automated region of interest smart selective Raman mapping.
