Array Microcell Method Coupled with Microfluidic Cell Traps for Single-cell Quantification

Description

Recently our lab presented the array microcell method (AMCM), where a droplet at the tip of a movable micropipette (inner diameter: 50 µm) functions as an electrochemical cell, in which the electrode area is defined by a microelectrode on a microelectrode array (MEA). A key attribute of AMCM is localized electrochemistry can be individually studied at each microelectrode. By combining a MEA with additional microfluidic trapping features, single cells can be sorted into microelectrodes. Here, we present a glucose oxidase (GOx) functionalized carbon MEA, integrated with microfluidic features to measure the intra-cellular glucose concentration of single human red blood cells (RBCs). GOx functionalization was performed via diazonium chemistry and EDS/NHS coupling on pyrolyzed photoresist films-MEAs (PPF-MEAs). Glucose concentration was determined via redox mediator in phosphate buffered saline (PBS) by chronoamperometry.
