Modulation of Ion Transport Across Nanopores

Ion channels are an example of a biological nanopore system which regulates ion transport through a variety of mechanisms. Inspired by an ion channel’s ability to modulate ion current, a synthetic nanopore system will be presented. A porous silicon nitride (SiNx) membrane was prepared by milling nanopores into the SiNx and applying a coating of Nafion to one side of the membrane to induce a negative surface charge. Bulk current-voltage measurements demonstrated that the Nafion-coated porous membrane displays ion current rectification at one voltage polarity versus the opposite polarity. Various modes of scanning ion conductance microscopy (SICM) were implemented to study the ion transport properties across individual pores. The advantage of SICM is the dimension of the nanopipette which allows the concentration within hundreds of nanometers of the nanopore to be probed and explored. Local ion transport across Nafion-coated pore was monitored as different potentials were applied across the membrane leading to ion accumulation and depletion. In the future, this synthetic nanopore system will be used to develop a more complex device to regulate ion transport.
